EZ Random Mutagenesis Kit는 PCR 기법을 활용하여 무작위 돌연변이를 원하는 확률로 생성하는 제품으로 PCR 반응에서 MnSO4와 dGTP를 이용하여 돌연변이를 조절할 수 있습니다. 돌연변이 발생율을 증가시키기 위해 dGTP의 농도를(100 μM) 일정하게 유지하면서 MnSO4농도를 (최대 100 μM) 증가시키면 됩니다. MnSO4를 최대 농도에서 (100 μM) 고정한 상태에서 dGTP의 농도를 (최대 300 μM) 증가 시키면 더 높은 돌연변이율이 발생합니다. 다양한 버퍼 조건으로 무작위 돌연변이 발생은 1,000 bp당 최대 19개를 발생시킬 수 있습니다.

Standard reaction using 1 ng of plasmid template, 10 pmol of each primer and 25 cycles of PCR. All procedures completed in same thermocycler and analyzed by NGS. When the amount of dGTP and Mn2+ increases the error rate follows the increment.
